The green microalga Chlamydomonas reinhardtii was transitioned from conventional culturing methods in liquid medium to a spatially structured three-dimensional environment. This transition caused several morphological changes. Here, we show one of them, the development of a double-layered cell wall. Using structured illumination microscopy (SIM) on a Zeiss Elyra 7, we visualized the cell wall stained with concanavalin A in the green channel. The purple channel highlights the autofluorescence of chlorophyll within the U-shaped chloroplast of the alga, providing a detailed, high-resolution view of the alga’s transition.
