This micrograph was taken during inverted epifluorescence microscopy (microscope: Leica DMI 3000 B; camera: Leica DFC 490), using a fluorescence brightener. The analysis has revealed much more than originally intended. The “spiral” represents a chain of auxospores formed by the diatom Achnanthes taeniata (spring bloom, Baltic Sea). To my surprise, the brightener visualized some parts between the auxospores, apparently containing cellulose, carboxylated polysaccharides and/or callose, to which it strongly binds to. A lorica of the chrysophyte Dinobryon faculiferum was also stained and virtually invisible without exciting the brightener. The title of this entry was inspired by Florence + the Machine.
